Sirolimus Tablets are immunosuppressant medications primarily used to prevent organ transplant rejection and to treat certain rare diseases involving immune dysregulation. Sirolimus works by inhibiting the mTOR (mammalian target of rapamycin) pathway, thereby suppressing immune activation and reducing the risk of graft rejection. It is also used in specific cases such as lymphangioleiomyomatosis (LAM) to improve lung function and slow disease progression.
Sirolimus should be taken once daily, consistently with or without food, exactly as prescribed by a healthcare provider. Blood levels often need to be monitored to maintain safe and effective dosing.
Uses of Sirolimus Tablets
Transplant Medicine
Prevention of organ transplant rejection (primarily kidney transplants)
Reduction of immune-system activity to protect transplanted organs
Other Medical Uses
Management of lymphangioleiomyomatosis (LAM)
Treatment of certain immune-related or hematologic conditions (as directed by specialists)
Benefits of Sirolimus Tablets
Effective immunosuppression to prevent transplant rejection
mTOR inhibition helps regulate abnormal immune and cell-growth activity
Can be combined with other immunosuppressive agents for enhanced protection
Demonstrated benefit in slowing progression of LAM
Once-daily dosing improves convenience and adherence
How Sirolimus Works
Sirolimus is an mTOR inhibitor that reduces immune cell activity.
Mechanism of Action
Inhibits T-cell activation and proliferation: Prevents immune cells from attacking transplanted organs
Blocks mTOR pathway: Slows abnormal cell growth and inflammation
Reduces antibody-mediated responses: Supports long-term graft survival
This immunomodulatory effect lowers the risk of organ rejection and helps manage diseases involving excess immune activation or abnormal cell growth.
Side Effects
Common side effects

Diarrhea, nausea, or abdominal pain
High cholesterol or triglycerides
Skin rashes or acne
Joint or muscle pain
Swelling in the legs or ankles
Serious but less common side effects

Monitoring
Routine blood level checks
Lipid profile and kidney/liver function tests
Regular evaluation for infections
General Advice
